Paper-based Scaffold for Stem Cell Culture and Bone Regeneration |

Stem cell behaviors are highly regulated by various biochemical and biophysical cues from cell-surrounding microenvironments. Here we report a new surface engineered-paper scaffold system by initiated chemical vapor deposition (iCVD) process. iCVD is a one-step, solvent-free method to coat functional polymer films conformally onto various substrates. A waterproof and cell-adhesive polymer film was serially coated on the micro-structured surface of paper. The surface modified paper scaffold system demonstrated excellent biocompatibility and improved mechanical robustness. It is also suitable for cell adhesion, proliferation, and gene transfection. Moreover, the developed paper scaffold system could facilitate enhanced in vitro osteogenic differentiation of human adipose derived stem cells (hADSCs). Transplantation of hADSCs cultured on paper scaffolds significantly promoted in vivo bone formation in critical-sized calvarial bone defects. |
