Theranostics is a novel concept that combines therapeutics with diagnostics. Recently, theranostic nanoparticles have been developed to diagnose patient’s cancer at early-stage, as well as to treat the cancer by targeted drug therapy based on the diagnosis results. In this study, hyaluronic acid (HA) was chemically modified using the hydrophobic moiety, 5β-cholanic acid, and the surface of HA conjugates was pegylated. Owing to their amphiphilic characteristics, the resulting conjugates were capable of forming the self-assembled nanoparticles. In particular, a hydrophobic anticancer drug, camptothecin(CPT), was efficiently loaded in the pegylated hyaluronic acid nanoparticle (P-HA-NPs), and NIR dye, Cy5.5 was attached to the surface of P-HA-NPs for cancer theragnostics. These multifunctional nanoparticles showed efficient tumor imaging ability as well as excellent therapeutic efficacy via tumor targeting ability of P-HA-NPs in the tumor-bearing mice. |
