제목 SustainableProduction of Squalene from Methane Using a Methane-Oxidizing Bacterium, Methylomonas sp. DH-1
초록 Methane is not only one of the powerful greenhousegas, but also it is considered as an inexpensive and abundant feedstock for sustainable manufacturing. Wereport the sustainable production of squalene frommethane using ametabolically engineered methanotroph, Methylomonassp. DH-1 strain. First, a base strain for the Cre-lox-based automatic genetic engineering was constructed for efficient strain development. Second, the base strain was metabolically engineeredto improve squalene production by deleting the pds-ald-crtN2 gene cluster and overexpressing the squalene synthase,which are responsible for increasing the pool ofsqualene precursors and carrying strong metabolic flux toward squalene. Finally, the strain produced 31.3mg/L of squalene from 3.09 g/L of methane with the yieldand productivity of 10.2 mg/g and 746.2 μg/L/h, respectively, by fed-batchfermentation.
