Today, the environmental pollution problem of plastics is a serious threat. As a result, enzymatic recycling methods are in the spotlight. Many promising enzymes that can degrade polyethylene terephthalate(PET) have been discovered. However, there are many difficulties in researching PET degradation enzymes because there is no high-throughput screening(HTS) system for mutant’s activity comparison. In this study, two screening methods were compared to detect terephthalic acid(TPA), the final degradation product of PET. First, a screening method was constructed in which TPA was reacted with an hydroxyl radical to change it into a fluorescent material. Second, GFP biosensor methods using activator discovered from Comamonas testosteroni and Escherichia coli were studied. Through this study, we tried to make HTS system for TPA detection. |
