Metabolic activity of encapsulated HepG2 cell spheroids according to the mechanical property of the gelatin hydrogel |

Hydrogels are used as cell culture scaffolds both in vivo and in vitro because they provide an environment similar to a living body due to their high water content and flexibility. In addition, the hydrogel can serve as a matrix in spheroid fabrication. This experiment shows that the activity of spheroids varies according to physicochemical properties such as mechanical properties and swelling rate. The physical properties of the hydrogel were measured using rheology, and MTT and Live/Dead assay were performed to confirm the reaction when a hydrogel with different properties was used as a scaffold for hepg2 spheroid culture. Hepg2 activity was confirmed by quantifying the concentrations of urea and albumin produced in the supernatant using urea and albumin assays, respectively. It is expected that this will help in the development of biomimetic liver systems and modeling to improve the predictive sensitivity to hepatotoxic drugs. |
