A Versatile Nanoconstruct Harnessing Tumor Retention, Permeation and Activatable Drug Release properties for Peritoneal Carcinomatosis treatment |

Poor prognosis and low survival rate among peritoneal carcinomatosis (PC) patients prompted us to design a special drug delivery systems with high anti tumor activity and survival rate. Herein, we designed anti-cancer drug loaded glutathione responsive micelles that can enhance peritoneal retention time, penetrate tumor nodes and could release the drug in glutathione rich environment of cancer cells. Nanoparticles showed enhanced cellular uptake, cancer cell toxicity both in vitro and 3D tumor spheroids studies. Nanoparticles administered via intraperitoneal route demonstrated prolonged peritoneal retention and low systemic. Also, intraperitoneal chemotherapy showed significant inhibition of tumor nodes. Immunohistochemistry of tumor tissue demonstrated the high apoptosis and tissue damage. Overall, the design of drug loaded nanoparticles which could increase the peritoneal retention and tumor penetration proved to be an effective strategy for treating PC. |
