Immunogenic cell death (ICD) is crucial for effective cancer immunotherapy. Recently, the ICD induced by electrical ablation (EA) was revealed. The use of EA has the advantage of causing minor damage to the tissue surrounding the target site than conventional thermal ablation. This study evaluates EA-mediated ICD in vitro and in vivo to optimize electrical voltage and pulse conditions for effective ICD induction. The viability of cancer cells cultured in 2D and 3D conditions was measured after applying EA under various voltage and pulse conditions. We also analyzed the expression of damage-associated molecular pattern markers of ICD induced by EA. Furthermore, in in vivo experiments with tumor models, the therapeutic efficacy and survival rate after EA treatment were measured to identify the optimal conditions. Taken all together, we believed that the new approach for induction ICD by EA could be a good option for effective cancer immunotherapy. |
