Development of Ionizable lipid nanoparticles and their formulation enables the liver cell type specific delivery of RNA therapeutics for gene regulation |

Lipid nanoparticles (LNPs) are one of the efficient methods for delivering RNA therapeutics to hepatocyte. Gene regulation potency of RNA therapeutics are highly dependent on LNP formulations. Generally, various formulations of LNPs have been focused on targeting primary hepatocytes. However, LNPs mediated liver cell type specific delivery for RNA therapeutics has not been studied well. In this study, we prepared amine-epoxide based ionizable lipids. To prepare amine-epoxide based ionizable lipid, we utilized amines based on piperazine, piperidine. By our newly developed amine-epoxide based LNPs, we were able to deliver RNA therapeutics to the different types of liver cells by changing the amount of polyethylene glycol (PEG)-lipid in the LNP formulation. This approach allows size dependent liver cell type specific uptake of LNPs. |
