제목 Optimization of lipid nanoparticles (LNPs) formulation for the liver specific delivery of gaussian luciferase (gLuc) mRNA ​
초록 Lipid nanoparticles have received a lot of attention for the delivery of nucleic acid since the COVID pandemic. In the evaluation of LNP formulation, secreted reporter proteins are important for detection and  expression of mRNA. Firefly luciferase mRNA is often used for the evaluation of such phenomenon; however, it has a disadvantage in that the level of protein expression can be detected only organs, not in blood. The Gaussian luciferase reporter has considerable advantages against other reporter gene. It is more sensitive than other  reporters and can be quickly detected in blood, urine as well as organs. To facilitate the monitoring of in vivo delivery of Gluc mRNA, we optimized the LNP formulation. To examine Gluc expression profile, we injected LNP containing mRNA encoding Gaussian gene into mice by i.v injection and blood was withdrawn at different time points. These results suggest that Gluc mRNA- LNP platform may provide a useful tool to evaluate in vivo protein expression.
