The aim of this study is to investigate the potential use of layer-by-layer (LbL) coating technique to control the release rate of oxide (NO) from donor liposomes coating a metal stent. The NO release profiles showed very promising results and the NO release rate was sustained up to 5 days. It was demonstrated that endothelial cell (EC) viability was maintained for around 5 days, while smooth muscle cells (SMCs) were suppressed. It was also found that fibrinogen deposition in vitro was significantly decreased. Finally, in vivo study using a porcine coronary injury model showed that our surface modifications enhanced therapeutic effects in all parameters except fibrin score. In addition, morphometric SEM images of the lumen of arteries with implanted stents showed significant differences in the lumen coverage, indicating enhanced endothelialization. |
