Development of an in vitro co-culture device for the investigation of host–microbe interactions through LC-MS/MS based multiomics approaches |

Akkermansia muciniphila is one of the next-generation probiotics for promoting human health. But the use of it as a cell therapy requires studying the molecular-level mechanisms of how it affects human intestinal epithelial cells. Here, we developed a Mimetic Intestinal Host-Microbe Interaction Coculture System (MIMICS) suitable for multiomics studies. And the physiological changes of intestinal epithelial cells due to co-culture with active A. muciniphila were analyzed. First, it was confirmed that the physiological activities of the two cells were stably maintained. Second, the membrane integrity of Caco-2 cells is maintained and up-regulation of tight junction proteins. Third, Caco-2 cells induced enhancement of energy production through activation of lipid metabolism. Consequently, we propose a novel mechanism for effects caused by A. muciniphila and MIMICS will be very useful for studies investigating the effects of specific gut microbiota at the molecular level. |
