제목 Ex vivo natural killer cell surfaces modulation by polymeric cellular sticker for augmented antitumor efficacy
초록 Natural killer (NK) cells are a specialized immune effector. Here, we designed multifunctional cell surface coating materials. This polymeric cellular sticker contains 6-major functional moieties: (1) surface anchoring lipid, (2) PEG blocker, (3) positively charged moiety, (4) fluorescence dye, (5) cancer recognition moiety, and (6) stimuli responsive degradable linker. NK cell surface was homogeneously coated and remained for 48 hr without affecting the intrinsic properties of the coated NK cells. Anticancer efficacy of coated NK cells was enhanced by receptor-ligand binding. Anti-folate was released from the sticker in response to the molecules leaked by cancer disruption, thereby increasing the anticancer efficacy secondarily. Moreover, coated NK cells exhibited facilitated tumor tissue infiltration and tumor apoptosis in MDA-MB-231 mouse xenograft model. Therefore, multifunctional NK cell coating materials could be utilized as an efficient anti-cancer immunotherapy platform.
