Promising approach for improving the coating stability and in vivo performance of biodegradable polymer-coated sirolimus-eluting stent |
초록 |
The stability of drug-in-polymer coating on the stent surface plays an important role in determining the performance of drug-eluting stent (DES). DES still suffers from coating delamination that could lead to non-uniform local drug distribution, restenosis and thrombosis. In this study, a thin layer of poly(lactide) brushes were introduced on the stent surface and a matrix of PDLLA & sirolimus were coated on the unmodified and brush modified stents. The drug release from the brush modified stent showed a sustained release rate as compared to unmodified one. The SEM images of the drug-in-polymer matrix coating after 2 months displayed coating peeling and delamination from unmodified stent as compared to the brush modified stents. Moreover, the in vivo study of brush modified stent indicated significant decrease in restenosis area as compared to unmodified stent. Therefore, the introduction of nano-brushes on a stent surface could promisingly improve the in vivo performance of DES. |
