제목 Sequential delivery system of polymeric siRNA nanocomplex via rolling circle transcription for cancer therapy.
초록 siRNA has been received a great attention as emerging gene therapeutics due to its superior capacity of specific gene silencing on post-transcription level. However, the clinical application of siRNA is still limited due to its inherent properties and delivery problems. Herein, we developed the polymeric siRNA via rolling circle transcription process, and delivered the polymeric siRNA to tumor by using redox-sensitive glycol chitosan (GC) derivative as a gene carrier. The novel designed polymeric siRNA nanocomplex (PRC) showed the highly enhanced stability and less cytotoxicity, compared to conventional polyelectrolyte complexes. Because the PRC was designed to release active siRNA in cytoplasmic region, they showed high gene silencing efficacy with efficacy with efficient tumor targeting ability. According to the results, our PRC can be considered as a platform technology for efficient tumor targeted siRNA delivery.
