Polymer-Templated Protein Nanoball (PTPNB) Based on Poly(Lactic Acid): A Promising System for Drug Delivery |

The development of protein-polymer nanostructures has become a critical interest in the medical and biological fields due to versatile functionalities and special properties of proteins. In this regard, we describe a simple one-pot procedure for producing PTPNB consisting of PLA core and His-tagged GFP as shell. PLA was selected as hydrophobic polymer template due to their reduced toxicity and ability to clear through biological systems. PTPNB was prepared by simple mixing of Ni-NTA-end-functionalized PLA dissolved in DMF and His-tagged GFP solution dissolved in phosphate buffer solution. In a first to assembling as PTPNB, PLA aggregates due to its poor solubility in water, while their temporary stabilization transpires through hydrophilic Ni-NTA. Then, GFP conjugates with PLA particle through specific interaction between histidine tag and Ni-NTA complexation. The successful formation of PTPNB was confirmed by various characterization techniques such as TEM, DLS and AFM. |
