제목 | Self-sustaining oxygen gradient formation and its use for investigating the gut bacteria/host interactions |
초록 | Oxygen is one of the most important signaling molecules in cellular functions. The human intestine has a distinct oxygen profile across the tissue with the oxygen-depleted lumen and highly oxygenated intestinal tissues through extensive vasculatures, which imposes a great challenge in creating an in vitro model system. Mimicking this oxygen gradient across the intestinal tissue is critical for recreating the ecosystem of the intestine with gut microbes that have significant influences on the health and disease states of the host. We developed an in vitro intestinal model system that forms a self-sustaining oxygen gradient exploiting cellular oxygen consumption without any external pump or tubing for deoxygenation. Multiple gut bacterial strains were successfully cocultured with the primary human intestinal epithelial cells in this model system. This model system offers a valuable platform for investigative studies on intestinal physiology and testing/screening applications. |
