Hydrogel-based SPR sensor for detection of cancer biomarkers via multivalent protein binding |

Interleukin-2 (IL-2) and its soluble receptor, IL-2α, are common biomarkers for cancer and autoimmune diseases. Conventionally, the evaluation of these biomarkers has been conducted with the enzyme-linked immunosorbent assay (ELISA) due to the precision afforded. However, ELISA is time-consuming and require many complicated steps. Herein, we report a new tool for rapid and accurate cancer diagnosis using untreated serum. pNIPAm-co-AAc hydrogels conjugated with IL-2Rꞵ and IL-2Rγ, were utilized to form sensor chips. In presence of IL-2 and sIL-2Rα, tetrameric complexes form and cause a multivalent protein binding (MPB) effect, which is then observed via surface plasmon resonance (SPR). We show that cancer diagnosis can be conducted with 1000x diluted serum in 10 minutes and our results were well correlated with results obtained from ELISA. Our finding indicates that MPB-based cancer diagnosis can be used as a tool to identify a target molecule in unpurified solutions. |
