Development of dual-functional nanofibrous scaffolds for skin tissue engineering |

Recently, there are increasing social and ethical concerns regarding the use of animal test for property analysis of new chemicals, novel system for replacing animal testing model has been pursued in tissue engineering field. In this study, we developed PLCL nanofibrous scaffolds, and then AAc was grafted to introduce carboxylic acid group using r-ray irradiation. To increase biological function of the scaffolds, EGF and gelatin were further immobilized on the scaffolds. The properties of the scaffolds were analyzed by SEM, TBO uptake, ATR-FTIR, water contact angle measurement, ELISA and fluorescamine assay. During the 10 days of hMSC culture, adhesion, spreading, and proliferation were significantly increased on the dual-functional scaffolds, we also confirmed that differentiation to epidermal lineage was up-regulated on the EG-AP scaffolds. Therefore, our results suggest that dual-functional scaffolds may be presented as an alternative tool in skin tissue engineering. |
