제목 | Design of Extracellular Matrix for Liver Tissue Engineering |
초록 | The cellular environment is dominated by the extracellular matrix (ECM), which is mainly composed of collagens, proteoglycans, and glycosaminoglycans (GAGs) that are secreted and assembled locally into an organized network to which cells attach. Interest in the ECM has increased dramatically in recent years due to an increasing awareness of its relevance to the regulation of cell growth, morphology, and differentiation. Heparin as one of species of the GAGs, is anionic polysaccharide and can bind tightly the GAGs integration site and become a “bridge” between cells and materials [1]. Chitosan as a binary heteropolysaccharide containing [1-4] linked 2-acetamide-2-deoxy-ß-D-glucopyranose and 2-amino-2-deoxy-ß-D-glucopyranose residues has characteristics similar to GAGs and is biodegradable and non-toxic. In this study, galactose moiety was covalently coupled with chitosan for hepatocytes attachment and then, heparin was immobilized into the galactosylated chitosan (GC) to enhance hepatocytes functions.
