To efficient gene delivery, we synthesized thermosensitive gene carrier, low molecular weight water-soluble chitosan(LMWSC)-g-poly(N-isopropylacrylamide)(PNIPAAm)(CSPNP). Degree of substitution(DS) and molecular weight of CSPNP were determined by 1H NMR and GPC. The thermosensitive behavior of CSPNP was confirmed by turbidity using UV-VIS spectrometer and 1H NMR. We prepared to CSPNP/DNA complex using ionic interaction between CSPNP and naked DNA for gene carrier. The complex size was dramatically reduced(700 nm→50 nm) with temperature increment upon LCST of CSPNP(37℃). The in vitro gene transfection revealed that CSPNP95, which has high gene transfection, may be considered a potential candidate for efficient non-viral gene carier. |
