Dual pH/Ultrasound-Responsive Sonotheranostic Extracellular Vesicles for Efficient Chemo-sonodynamic Cancertherapy and Photoacoustic Imaging |
초록 |
Sonodynamic therapy (SDT) is a new therapeutic for noninvasive cancer treatment using ultrasound (US) to penetrate and activate sonosensitizers. Extracellular vesicles (EVs) were employed as biocompatible nanocarriers for drug delivery. Herein, EVs were utilized as dual stimuli-responsive carriers to deliver indocyanine green (ICG)(sonosensitizer/photoacoustic imaging agent), paclitaxe l(PTX)(chemo drug), sodium bicarbonate (SBC)(pH-responsive) into tumor cells. The endo/lysosomes acid pH environment bursted EV membrane by CO2 bubbles generation from the SBC and facilitated the release of ICG and PTX, and it was increasing when treated with US. SBC-EV(ICG/PTX) exhibited efficient anticancer activity against breast tumor cells and the accumulation of SBC-EV(ICG/PTX) in tumor-bearing mice was demonstrated by high-resolution PA imaging. SBC-EV(ICG/PTX) are promising sonotheranostic nanoplatforms for safe and efficient photoacoustic imaging and chemo-sonodynamic combination cancer therapy. |
