The PEAD, polycation was widely used to prepare coacervate with the anionic polymer heparin. However, coacervate have difficulty in their colloidal stability. Herein, we investigated the effect of PEGylation of PEAD (mP-PEAD) on the stability of the coacervate under aqueous conditions. A series of mP-PEAD (mP350-PEAD, mP750-PEAD, mP2000-PEAD) were synthesized. As a result, the PEGylation with lower Mw resulted in more positive to surface charge. The mP350_Coa maintained their size over time and also exhibited improving their stability in presence of competing ion (i.e., 0.15 M NaCl). In addition, VEGF 165 were incorporated in our platform with high loading efficiency (98-99 %) and showed a sustained release pattern up to 21 days. Moreover, mP350_Coa significantly increased tube formation due to relatively rapid release of VEGF 165. Overall, we expected our developed platform with mP350 could be ideal strategy for efficiently delivering therapeutic protein. |
