Synthesis of hyaluronic acid based protease degradable hydrogel |

Degradable hydrogels are an important advanced materials in the field of biomedical applications for the purposes of tissue engineering and drug delivery. This presentation describes a method for preparing hydrogels degradable by alpha-chymotrypsin. The protease degradable hydrogel is composed of acrylated hyaluronic acid crosslinked by a peptide crosslinker containing a proteolytic degradation site. Hyaluronic acid was used because of enhanced biocompatibility and CD44 targeting for cancer therapy. Proteolytic degradation site is a peptide that sequence ‘CYKC’ reacted by alpha-chymotrypsin. Two thiol groups from terminal cysteines can be crosslinked with acrylated hyaluronic acid by thiol-ene click reaction. We envision that these protease degradable hydrogels are useful for tissue engineering and drug delivery. |
