One of the polyphenol molecules from nature, catechin, has biochemical properties such as anti-angiogenesis, anti-inflammatory, and anti-oxidant. Corneal treatment requires such properties for clear vision and healthy ocular lens. Despite of advantageous properties of polyphenol, it has not been used in corneal treatment due to very low solubility for being used as eyedrop-based formulation. In this study, we enhanced the solubility of catechin in water through complex formation with poly(ethylene glycol) (PEG). Catechin dissolved methanol solution and PEG dissolved aqueous solution are mixed and then freeze-dried which results in physical complex of catechin/PEG. The solubility of catechin:PEG complex is increased five hundred times (50mg/ml) while the anti-vascularization remained effective. Various weight ratios of catechin:PEG have been investigated for their solubility. The optimal weight ratio is determined to be 1:5 with particle size of about 100~200 nm. In vivo study on damaged rabbit corneal epithelium indicated that catechin has anti-angiogenic, anti-inflammatory effects. |
