Genetically Engineered Polypeptide-based Nanostructures for Theragnosis |

Polymeric nanostructures for theragnosis were developed to simultaneously detect and treat a disease. Especially, imaging and inhibition of neovascularization induced by uncontrolled angiogenesis are of interest to treat various diseases. We report herein the self-assembled nanostructures of genetically engineered fusion proteins, which were composed of penta-helices for the formation of coiled-coils, elastin-based polypeptide with stimuli-responsiveness for non-chromatographic purification tags and membrane-receptor targeting peptides for anti-angiogenesis. Functional peptides of the nanostructures were exposed to the aqueous environments, facilitating specific molecular interactions against the receptors and inhibition of neovascularization, as confirmed by enzyme-linked immunosorbent assay and fluorescence imaging via tubing assay of human umbilical vein endothelial cells. Conclusively, these self-assembled nanostructures would be potential as all-in-one theragnostic nanostructures. |
