NON-VIRAL DIRECT CONVERSION OF PRIMARY FIBROBLASTS INTO NEURONAL CELLS USING BIODEGRADABLE POLYMER NANOPARTICLES |

Recent studies have shown that defined sets of transcription factors can directly reprogram differentiated cells into another linage without passing through a pluripotent stem cell-like state. Most studies reported so far have used viral vectors to deliver the genes, which causes the safety issues, hence limits the scope of their potential clinical applications. Here, we report a novel method for reprogramming mouse fibroblasts directly into neuronal cells using specific biodegradable polymer nanoparticles. These induced neuronal cells uniformly displayed morphological and molecular features of neuronal cells, and exhibited electrophysiological functions. Our non-viral direct conversion approach may endorse the prospect of autologous cell therapy for neurodegenerative disorder.
