제목 | Gene transfection via Small molecular conjugates with Energy-independent processes |
초록 | Cell penetrating peptides (CPPs), a series of cationic that exert macromolecular transduction across the membrane were identified. The reason for the use of CPPs for intracellular delivery of small molecules, proteins, and nucleic acid drugs is that it seems to mediate transfer of macromolecules across the cell membrane endocytosis-dependent processes. A class of bile acids (BAs) such as deoxycholic acid (DA) has an unusual structural feature in which two to three hydrophilic hydroxyl groups face to the same direction and hydrophobic steroidal hydrocarbons orient to the opposite direction. The structural feature is similar to the CPPs which may play an important role for the membrane fusion or permeabilization of the peptide. Herein, a molecular amphiphile (BA) and a cationic polymer, polyethylenimine(PEI), one of themost widely used gene carriers were conjugated to mime the CPPs. |
