Development and evaluation of novel cosmetic materials promoting cell-cell interaction and junction protein activation |

We have examined the effect of several synthetic peptides derived from the cell-adhesive sequence cell-cell interactions using keratinocytes. Gardekera, a synthetic peptide consisting of 18 amino acids induced to promote cell-cell interaction of keratinocytes. This peptide also increased the expression of tight junction proteins such as claudin family and occludin. The transcellular electrical resistance (TER) increase was accompanied by a decrease in paracellular permeability in peptide-treated cells. We also examined an approach to assess the skin irritation potential of this peptide using EpiDerm reconstructed with human skin tissue; however, cytotoxicity was not observed for peptide-treated group. Peptide treatment revealed increased thickness of basal, spinous, granular and corneum compared to the control group. Notably, granular layer with expression of tight junction proteins appeared to rise into considerably thicker and more clear. |
