제목 장내미생물 공배양을 위한 대장 모델 개발 Development of an in vitro human colon model system for gut bacterial coculture
초록 Interactions of gut microbes with their host have significant impacts on the health and disease states of the host. Understanding these interactions can lead to discovering new diagnostic tools and therapeutic targets. An in vitro gut bacterial co-culture model system with human colon cells can be a valuable tool for testing hypotheses on gut microbes/host interactions and screening probiotics or pathogens with much less cost than animal models. However, creating a physiological oxygen gradient necessary for survivals of human cells and gut bacteria, primarily anaerobic, within one system has been challenging. We developed a novel and simple in vitro model system with a physiological oxygen gradient using cellular respiration and modulating oxygen influx. Multiple anaerobic gut bacteria were successfully co-cultured with the human colon epithelial cells. This platform provides a valuable platform for investigating and further utilizing the interactions of gut bacteria with human cells.
