제목 Nanoassembly of chemically modified Cas9 ribonucleoprotein for in vivo gene editing and cancer immunotherapy
초록 Gene therapy using clustered regular interspaced short palindromic repeats (CRISPR) system is challenging because of poor delivery and efficacy in vivo. We report the development of a nano-assembled ribonucleoprotein complex (NanoRNP) that can efficiently block the PD-L1 immune checkpoint in vivo and induce anti-tumor effects. Chemically modified CRISPR-associated protein 9 (Cas9) with single guide RNA and modified DNA oligonucleotide formed a stabilized NanoRNP. Delivery of NanoRNP in vivo in a mouse melanoma model can induce indels in tumor, thus significantly inhibiting tumor growth without including combination therapy. Blockade of the PD-L1 checkpoint by NanoRNP in tumor promotes suppression of immunosuppressive myeloid cells, as well as T cell infiltration and effector cytokine release, which are hallmarks of antitumor immune activation. This system proposes a promising strategy as an in vivo gene editing platform for cancer immunotherapy.
