Chemo-photodynamic combination therapy has been considered an excellent strategy for various cancer types. However, there are limitations such as abnormal increase in tumor angiogenesis by reactive oxygen species generated by photodynamic therapy and degradation of genetic materials delivered with photosensitizer. In this study, we developed the complex of doxorubicin-encapsulating anti-angiogenic siRNA nanoparticle and Ce6-encapsulating microbubble to overcome limitations of existing therapies. The first part, the Ce6-encapsulating microbubble, carries the photosensitizer and is a trigger for local delivery system. The other part, doxorubicin-encapsulating siRNA nanoparticles, was coated with two biopolymers to prevent damage of the genetic materials. Both in vitro and in vivo results of the experiments showed successful inhibition of angiogenesis with minimized siRNA damage by ROS as well as improving therapeutic effect by combination therapy inducing local delivery by ultrasound. |
