제목 | Changeable polymers in the tumor microenvironment for effective tumor immunotherapy |
초록 | Tumor immunotherapyis encouraging tumor treatment that can substitute the treatment such assurgery, chemo drug. However, in the tumor microenvironment (TME), itstherapeutic effect is limited. To resolve this problem, we designed the nanoparticleswhose properties change at TME. We conjugated an acid-sensitive molecule to acationic polymer and loaded PS (ACPP). In the TME, acidic sensitive molecules detachand the charge of the particle changes. ACPP amasses in the tumor and induces tumordeath. Generated tumor antigens induce recruitment of cytotoxic T cells and thematuration of dendritic cells. We confirmed the pH sensitive propertiesthrough zeta-potential and dynamic laser scattering. In vitro, thecytotoxicity and membrane disruption in an acidic environment were confirmed byMTT assay, confocal microscopy. In vivo, anti-tumor and immunotherapeuticeffects are confirmed by the flow cytometry. Biocompatibility was affirmedH&E staining of tissue and serum chemistry levels. |
