Stem cells with the capacity to self–renew and differentiate into specific cell types have been considered as a promising cell source for tissue engineering and disease modeling. However, development of cell culture system for reliable, reproducible, scalable production of the stem cells is remained as a challenge for scientists. In recent, a variety of cell culture systems based on embryonic fibroblast feeder, matrigel, hyaluronic acid are adapted for stem cell proliferation. However, the contamination by animal-derived cell/pathogen transmission, difficult cell harvesting are considered as drawbacks for clinical applications. In this study, we developed a 3D cell culture system consisted of viscous hyaluronic acid(HA) solution and porous membrane with selective permeability(permeation of nutrients/O2, but prevention of HA leakage) for stem cell proliferation. The morphology of membrane, permeability test, and cell proliferation behavior in the 3D culture system were investigated. |
