Polymerase chain reaction (PCR) is considered as a golden standard in molecular diagnostics for a multitude of applications ranging from precision medicine to point-of-care testing. However, adequate amplification of DNA for conventional PCR requires several hours due to large reaction volumes of several tens of microliters. Therefore, a novel PCR technique for the rapid amplification is highly desirable for early diagnosis of infectious diseases such as COVID-19. Here, we propose water-wettable void plasmonic nanogap between metal nanoparticle and film as a novel platform for the rapid amplification and optical detection of DNA. The void nanogap is directly visualized by electron microscope. We successfully amplified DNA within few minutes based on photothermal conversion in the nanogap. Then, the amplified DNA is sensitively detected in situ via surface-enhanced Raman spectroscopy without labeling. |
