M13 bacteriophage (phage) is one of the rising biomaterials for detection because they are resistant to harsh condition such as high salt condition and acidic pH, and stable to storage. Moreover, Phage can display foreign peptides which bind to various target molecules through genetic engineering. In this study, we present a simple strategy for detecting Aspergillus niger (A. niger) spore by spore-binding phages. A. niger spore binding phages screened from M13 phage 12-mer peptide library. And then, we performed phage titration binding assay using the selected pool of six phage clones. After FITC labeling the phages, the interaction between fungal spore and spore-binding phages were also studied by flow cytometry assay. We believe that phage-based detection system has a promising approach to detecting diverse targets by altering phage sequence. |
