Novel Application of PCL-Gelatin Electrospun Membranes to Prevent Postoperative Leakage |

Anastomotic leakage due to suture line disruption after abdominal surgery is one of crucial factors affecting a patient’s survival. To support various kinds of clinical suture sites and prevent leakages, polycaprolactone (PCL)-gelatin electrospun membranes were fabricated with different ratios (1:1, 1:2, 1:3 and 1:4) of PCL to gelatin. Higher gelatin content in the membrane resulted in lower fiber diameter and contact angle, leading to more hydrophilic membrane that made it easier to be attached to the defect site. The degradation rate of each PCL-gelatin electrospun membrane was also faster when gelatin content was higher. Cell viability was assessed using L929 cells. Among different ratios, the ratio of PCL to gelatin at 1:4 (P1G4) had the best result, showing the highest cell viability at more than 80% (control cell viability: 100%). According to in vivo results, when defected and sutured site of rat caecum was covered with P1G4 membrane, the site was recovered without any leakage at two weeks after surgery. Therefore, PCL-gelatin membrane has potential to be applied as a leakage-preventing membrane that aid recovery after surgery. |
