Macrophages mediate adaptive and innate immunity responses in the human immune system. Among the macrophages, tumor-associated macrophages (TAMs) are influenced by the tumor micro-environment. Moreover, tumor cells can disturb the immune response of macrophages and induce immune evasion. The anti-phagocytic signal-regulated by anti-phagocytic proteins have been called 'don't eat me signal,' including sialic acid-binding immunoglobulin type lectin-10 (SIGLEC-10) and CD24 recently revealed immune checkpoint (ICP). In this study, we demonstrate that targeting a specific glycan on CD24 has potential as a factor of immune checkpoint inhibition. Sambucus nigra agglutinin (SNA), a sialic acid-binding lectin, shows CD24 blockade and enhanced phagocytosis of melanoma tumors. Furthermore, Lectin is conjugated with photothermal nanoparticles for applying photothermal therapy (PTT). We find that Lectin conjugated photothermal nanoparticles applied with near-infrared (NIR) augment tumor cell phagocytosis and demonstrate therapeutic potential in cancer immunotherapy. |
