제목 | Local gene delivery system using polyethylenimine and iodized oil |
초록 | The gene delivery system using non-viral vectors recently draws many attentions due to the safety problems of viral vectors. However, there are still some weaknesses of non-viral vectors, such as relatively low transfection efficiency and the inability to target gene expression to the specific cell. Cationic polymer has been shown as a promising carrier among the non-viral gene delivery systems. Compared with other non-viral gene delivery systems, especially liposome or cationic lipid systems, polymer-DNA complexes are generally more stable. One of the successful polymeric gene delivery carriers is the polyethyleneimine (PEI)-based carrier. The site-specific delivery could establish and maintain the local gene expression, either by preventing its efflux from the arterial wall or by using vehicles or agents that had the prolonged duration of action. In this study, we select polyethylenimine as a polymeric gene delivery carrier and Lipiodol as an oil phase to make water-in-oil emulsions. The local gene delivery system using PEI and Lipiodol was successfully delivered the DNA to target cells .
