Biomimetic co-culture system for reconstruction of microvascular system using electrospun fiber |

Blood vessel is essential process for tissue regeneration to deliver oxygen and nutrients and maintain tissue metabolism. Here we developed a technique to reconstruct microvascular system using polycaprolactone (PCL) and gelatin based nanofibrous structure and co-culture system. Using simple electrospinning process, we fabricated 3D mesh scaffold to support endothelial cells (ECs) sprouting along the electrospun nanofiber, and co-culture with adipose-derived mesenchymal stem cells (ADSCs) supporting greater sprouting of ECs. The vascular formation in our system showed more effective paracrine signal from ADSCs to assist vascular formation of ECs than influence of growth factor. Co-cultured ECs expressed higher level of vascular markers (PECAM-1, podocalyxin). We established a bioengineered 3D culture system to promote fast and functional vascular. Co-culture system in nanofibrous scaffold can be applied in the future for restoration of blood vessel. |
