Development of target molecule detection and capture system utilizing fluorogenic molecular net; polymer based DNA hydrogel |
초록 |
We propose fluorometric sensor system for target molecule capture/detection utilizing polymer based DNA hydrogel (PD-hydrogel). The fluorescent PD-hydrogel composed by acrylamide, bis-acrylamide and Acryd-DNA was prepared by radical polymerization and applied to detect target; DNA mismatch and small molecule as ATP. Through introducing fluorophore modified competitor DNA (Fl-cpDNA), target capture/detection utilizing PD-hydrogel was confirmed by both fluorescence decrement of PD-hydrogel and fluorescence increment of remained solution since the Fl-cpDNA in PD-hydrogel were detached and released into the remained solution. Our developed PD-hydrogel platform would be easily applied to detection and capture of target selectively, by fluorogenic method. |
