제목 Nerve Regeneration through Asymmetrically Porous Nerve Guide Conduit with Nerve Growth Factor Gradient
초록 The sufficient functional recovery of damaged peripheral nerves has been considered as one of the clinical challenges. A therapeutic aim after peripheral nerve injury is thus to re-establish connectivity by aiding in axonal regeneration. It is well known that the nerve regeneration in the body is mediated by gradients of bioactive molecules including nerve growth factor. In this study, we prepared an asymmetrically porous polycaprolactone (PCL)/Pluronic F127 membrane with NGF gradient. The nerve guide conduit (NGC) was fabricated by rolling the membrane. The NGF loading amount and release profile along the NGF gradient direction of the prepared membrane, and the neurite outgrowth behavior of PC12 cells on the NGF gradient membrane were investigated. The long gap (2 cm) peripheral nerve regeneration through the NGC with NGF gradient was compared with that of uniform NGF loading using a rat model (This work was supported by the National Research Foundation of Korea (NRF-2011-0000244).
