Vinylsulfoneconjugated HeparinPluronic Nanogels for Intracellular Drug Delivery |

The application of polymeric nanocarriers to cancer therapeutics is an area of significant research interest with good efficacy in the local chemotherapy, diagnosis of tumor cell. Our group has developed a potential nanocarrier, heparinCyaPluronicVS (HP) nanogels. In this system, the disulfide bond in HP nanogel may enable drugs to release by responding to glutathione in the intracellular region and prolonged circulation without dissociation. Besides, an amphiphilic block containing a VS group with high reactivity toward thiols has been considered as a conjugation site to conjugate with targeting ligand or imaging agent. As an in vitro result, a considerable amount of heparin was released from HP after GSH suddenly treatment of 5 mM at a final concentration. It indicates that HP nanogels were indeed degraded by GSH, thereby facilitating the release of heparin. These results suggest that HP nanogel may be a good candidate for nanocarriers in efficient intracellular delivery. |
