Stimuli-responsive multistage nanoparticles for deep tumor penetration of doxorubicin |

Heterogeneous drug distribution within tumor tissue is one of the major hurdles in improving overall therapeutic efficacy. To resolve this issue, we have developed stimuli-responsive multistage nanoparticles, named as transformable nanoparticles (TNPs), composed of pH-sensitive amphiphilic block copolymer, doxorubicin (DOX), and DOX-conjugated ultrasmall gold nanoparticles (DOX-NS). After systemic administration of DOX-loaded TNPs (DOX-TNPs), they were accumulated into the tumor interstitium followed by disassembly of polymeric nanoparticles responding to the mildly acidic tumor environment which triggers the release of DOX and DOX-NS. The released DOX was accumulated in the perivascular region and the DOX-NS could penetrate deeply into the inside of the tumor. After internalization of DOX-NS, DOX would be released by the cleavage of the ester bond. In this study, we successfully demonstrated a great potential of the multistage system of DOX-TNPs in anticancer drug delivery. |
