Harmonizing technique of protein and DNA has been much attained due to rapid evolution of protein engineering. However, it is still challenging to enhance its sensitivity and selectivity. Herein, an electrophoresis system based on functionalized hydrogel was used for advanced DNA/protein harmonization. The hydrogel contained NTA-functional groups to conjugate specific protein, and the UV-degradable crosslinker was introduced to selectively separate DNA from hydrogel system. Due to its homogeneous porosity of the hydrogel, the DNA library could be separated by electrophoresis depending on its size. The DNAs were expressed in hydrogel using in-vitro translation system. In addition, the expressed proteins were bound on the place where it was created due to the NTA-functional groups. Using the UV-degradable crosslinker, we could separate specific DNA which can express high functional protein, and the separated DNA could be matched with the high functional protein using DNA sequencing. |
