Ppa-iRGD-Q as a Depot Peptide for Effective Photodynamic Therapy |

One of the major issues in photodynamic therapy (PDT) is the controlled release of singlet oxygen (1O2) for effective treatment with minimal side effects. Herein, we describe a pyropheophorbide a (Ppa) conjugated cyclic iRGD peptide with a quencher at the other end (Ppa-iRGD-Q). Ppa-iRGD-Q peptide was designed to recognize cancer cells by the RGD motif, and the Ppa could be internalized into cancer cells via protease cleavage. Subcutaneous injection of Ppa-iRGD-Q resulted in the formation of a protease operated depot, which remained stable at the peritumoral environment allowing sustained release of the Ppa. Ppa was well preserved within the tumor allowing in vivo detection of the fluorescence signal of Ppa and also effective generation of singlet oxygen. We propose Ppa-iRGD-Q as a suitable PDT drug candidate for the controlled release of the photosensitizer into the tumor. |
