Small interfering RNAs (siRNAs) are full of promise for the genomic therapeutics because of their high sequence-specificity and inhibition-efficiency to the target cell in mRNA level. But they could be easily broken in the course of the delivery to the target cell and their anionic characteristics prevent the cellular uptake. Therefore we conjugated the poly(ethyleneglycol) (PEG) to the 5’ anti-sense strand of siRNA for protecting from the RNA degradation by the biodegradable-disulfide bond between PEG and siRNA. Moreover we used the fusogenic peptide, KALA, conjugated with PEG for the condensation agent of siRNA-PEG. In our study, siRNAs for Vascular Endothelial Growth Factor gene silencing were used for the anti-angiogenesis agent, are protected by PEG effect, and could be taken in the small particle form (200nm) by KALA-PEG. The KALA-PEG/siRNA-PEG complex showed that its anti-angiogenesis effect can be the predominant system for cancer therapy. |
