According to the previous real-time bioimaging study using quantum dots (QDots), hyaluronate (HA) derivatives with 35 mol% carboxyl group modification maintaining enough binding sites for HA receptors were mainly accumulated in the liver, while those with 68 mol% modification losing much of HA characteristics were evenly distributed to the tissues in the body. In this work, target specific long acting HA interferon α conjugate was successfully synthesized by the specific coupling reaction between aldehyde modified HA and N-terminal group of the protein. The interferon α content could be controlled in the range of 2~10 molecules per single HA chain with a conjugation efficiency higher than 95%. In vitro MTS test using Daudi cells confirmed the biological activity of the HA interferon α conjugate. The target specific HA interferon α conjugate will be investigated further for the treatment of liver cancer and cirrhosis. |
