Fabrication of microcapsules for sustained release of doxorubicin by O/W method |

In this study, we formulated PLGA/dextran double-layered microspheres by varying the molecular weight and concentration of PLGA and examined their feasibility as drug carriers for sustained drug release. Doxorubicin loaded dextran/PLGA microspheres were prepared by an oil/water emulsion method. The in vitro doxorubicin release from the double-layered microspheres was studied in phosphate buffered saline incubating the microspheres at 37℃ under constant shaking. The release profiles show that all double layered microspheres had a minimal initial burst. The drug release studies in vitro suggest that the release of hydrophilic drugs from the double layered microspheres is highly dependent on the molecular weight and concentration of PLGA. The results from this study indicate that dextran/PLGA double-layered microspheres by a simple method present great potential as drug carriers for sustained and controlled drug delivery.
