Recently, bone regeneration using tissue engineering technique combined with a local gene delivery system has been considered as one of the promising therapeutic methods. In this study, we prepared plasmid DNA (with BMP-2 encoding) complex-loaded polycaprolactone/Pluronic F127 asymmetrically porous membrane for more enhanced bone regeneration. For effective plasmid DNA release from the membrane and the following transfection of plasmid DNA into stem cells, the PCL membrane was hydrophilized by Pluronic F127 and the plasmid DNA was complexed with polyethyleneimine-polyethylene glycol. The pDNA/PEI-PEG complex was continuously released from the PCL/Pluronic F127 membrane for a long period of time (more than 3 months). The pDNA complex-loaded PCL/Pluronic F127 membrane showed faster bone regeneration than the control and membrane without pDNA. From the results, we could conclude that the pDNA/PEI-PEG complex-loaded GBR membrane may be a good candidate for effective bone regeneration. |
