Glucose-responsive polymeric nanoparticles for cancer therapy |

Functionalized polymeric nanoparticles has recently found potential for the detection and treatment of various diseases including cancer. Various cancer cells show critical damage of mitochondrial respiration and hypoxia stress, leading to high aerobic glycolysis compared to normal cells. In this study, we hypothesized that glucose-responsive nanoparticles could be useful to induce glucose deprivation of cancer cells and their metabolic control could lead to apoptotic cell death. Phenylboronic acid, reversibly binds to diols in glucose, was chemically conjugated to the backbone of polysaccharide and glucose-responsive nanoparticles were prepared via self-assembly. Various physiochemical properties of glucose-responsive nanoparticles and their anti-glycolysis effect were investigated in vitro. We also evaluated anti-cancer efficacy of glucose-responsive nanoparticles using a tumor-bearing mouse model after intravenous injection. |
